이번 발표 논문은 22년 5월 Cell Reports 에 출판된
'Germinal center expansion but not plasmablast differentiation is proportional to peptide-MHCII density via CD40-CD40L signaling strength' 입니다.
GC B cell 과 Tfh 의 interaction 은 humoral immune responses 에 있어서 가장 중요하다고 알려져 있습니다. GC B cell 과 Tfh 의 cognate 한 peptide-MHCII-TCR signal 을 통해 GC B cell 의 cell fate 가 조절된다고 알려져 있지만 아직 정확한 기전은 알려진 바가 없습니다.
본 연구에서는 aDEC205-OVA system 을 활용하여 B cell 이 uptake 한 antigen 을 follow 하여 T cell 과의 interaction 까지 확인할 수 있는 시스템을 구축하였으며,
GC B cell 이 cognate 한 Tfh 에 주는 MHCII signal 이 증가함 따라 GC B cell 의 proliferation 또한 증가하며, 이 interaction 의 key 는 CD40 strength 임을 밝혔습니다.
또한 이러한 signal 이 plasmablast differentiation 까지는 consistent 하게 영향을 주지 않는 것을 확인함으로서 GC B cell 에서 peptide-MHCII density 의 intrinsic role 을 밝힌 논문입니다.
