이번 주 발표자 도빈입니다.

이번 발표 논문은 올해 2월 Immunity 에 출판된 'Primary germinal center-resident T follicular helper cells are a physiologically distinct subset of CXCR5hiPD-1hi T follicular helper cells'입니다.

Follicular helper T cell (Tfh) 은 germinal center 에서 affinity maturation 에 관여하여 germinal center (GC) B cell 을 조절함으로서 antibody production 에 매우 중요하다고 알려져 있으며, phenotype 은 일반적으로 CXCR5hiPD-1hi 로 알려져 있습니다.

또한 위와 같은 CXCR5hiPD-1hi Tfh cell 들은 flow cytometry 로 분석하였을 경우 20% 정도를 차지한다고 알려져 잇는데, 실제 histology 상에서는 10%정도가 확인된다고 알려져 있습니다. 이와 같이 Tfh 는 보통 GC 내부 light zone 에 많이 분포하고 있지만 GC 외부에 존재한다는 것도 잘 알려져 있습니다.

하지만 기존에 GC 외부에 Tfh가 존재한다는 것은 알려져 있었지만, 어떠한 marker 로 이들을 구분해야하는지, 그렇다면 그 population 간에 어떠한 차이가 있는지는 알려진바가 없습니다.

본 연구팀은 CD90 과 S1pr2 가 resident / non-resident Tfh 가 구분할 수 있는 marker 라는 것을 확인주었으며, 이 population 이 서로 다른 development, transcriptional profile 을 가지는 distinct population 이라는 것을 보여주었습니다.
